The Influence of PLA-PEG on Zebrafish Complement System

摘要 通过实时定量PCR测定补体系统中主要补体因子的表达情况,探究了PLA-PEG(聚乳酸-聚乙二醇共聚物)在生物体内对免疫系统的影响。向成年斑马鱼腹腔注射PLA-PEG溶液,同时向对照组注射灭菌水,在注射后1,3,5,7,9,11d后分别取肝脏,提取RNA,反转录得到cDNA,使用RT-PCR仪器测定补体(C1r/s、C3、C4、C6、Bf、MASP、MBL)的表达量,并分析其在注射后的变化量。结果表明,注射后各补体因子表达量有不同程度的上调,说明PLA-PEG可以激活生物体内补体系统。 We discussed the immune process.of PLA-PEG(poly lactic acid polyethylene glycol copolymer)in vivo by determining the expression levels of complement factor by Real-time PCR.Method:Use the mature zebrafish as exprimental material,and intraperitoneal injection of PLA-PEG,while the control group is injected with sterile water.Take the liver of zebrafish at 1,3,5,7,9,11d post injection,then extract RNA and reverse transcriptase into cDNA.To determinate the expression levels of complement(C1r/s,C3,C4,C6,Bf,MBL,MASP) using Real-time PCR,we analyzed the change of expression levels after injection.The result showed all the complement factor expressing different levels increased after injection,and this could be considered the PLA-PEG can activate the immune response of complement system.
