摘要
目的:研究傣药"雅解沙把"对正常大鼠和CCL4致肝损伤大鼠肝药酶活性的影响。方法:傣药"雅解沙把"(2.43g/kg和0.27g/kg)对SD大鼠灌胃(ig)给药,每天1次,连续给药14d,分别测定细胞色素P450(CYP450)、细胞色素b5(Cytb5)的含量以及红霉素N-脱甲基酶(ERD)、氨基比林N-脱甲基酶(ADM)活性。结果:对正常大鼠,"雅解沙把"(2.43g/kg)能提高CYP450和Cyt b5的含量,诱导ERD和ADM活性;对CCL4致肝损伤大鼠,"雅解沙把"(2.43g/kg)能提高CYP450和Cyt b5的含量,对ERD和ADM活性无影响。结论:高剂量"雅解沙把"能提高正常大鼠和CCL4致肝损伤大鼠的CYP450、Cyt b5的含量,可以诱导正常大鼠的ERD和ADM活性,提高CYP3A和CYP2E1蛋白表达。
Objective:To investigate the effects of Yajieshaba on the activities of drug- metabolism enzymes in rat liver microsome. Methods:SD rats were given Yajieshaba (2.43,0.27g/kg) via i.g. gtt once a day for 14days. The contents of cytochrome P450, cytochrome b5 in rat liver microsome and the activity of ERD, ADM were assayed respectively. Results :For normal rats, Yajieshaba (2.43g/kg) can improve the content of CYP450 and Cyt b5, induces the activity of ERD and ADM;For liver injury induced by CC14 in rats, Yajieshaba (2.43g/ kg) can improve the content of CYP450 and Cyt b5, has no effect on the activity of ERD and ADM. Conclusion : High -dose administration of Yajieshaba (2.43g/kg) can improve the content of CYP450 and Cyt b5 for normal rats and liver injury induced by CC14 of rats, induces the activity of ERD and ADM, and increases protein ex- pression of CYP3A and CYP2E1.
