摘要
目的探讨变应性鼻炎(allergicrhinitis,AR)与干扰素调节因子5(interferonregulatoryfactor5,IRF5)的单核苷酸多态性(singlenucleotidepolymorphism,SNP)的相关性。方法采用聚合酶链反应和限制性片段长度多态性(polymerasechainreaction—restrictionfragmentlengthpolymorphism,PCR—RFLP)方法检测110例新加坡华人中AR患者及101例健康对照组人群的IRF5上的6个单核苷酸多态性位点(rs729302、rs4728142、rs3807306、rs2070197、rsl1770589、rs2280714),计算并比较两组人群基因型和等位基因频率。采用SPSS13.0软件进行统计学分析。结果IRF5基因的rs729302位点在人群中没有多态性,IRF5其他5个位点的基因型在AR组和健康对照组间差异均无统计学意义(X2值分别为0.21、5.02、0.01、2.91、0.37,P值均〉0.05);等位基因频率在AR组和健康对照组间差异均无统计学意义(X2值分别为0.00、2.78、0.01、2.31、0.00,P值均〉0.05)。结论IRF5基因多态性与AR无明显相关性。
Objective To study the possible association between interferon regulatory factor 5 (IRF5) gene polymorphism and allergic rhinitis (AR). Methods Six independent single nucleotide polymorphism (SNP, rs729302, rs4728142, rs3807306, rs2070197, rs11770589, rs2280714) were analyzed. The genotype and allele frequencies were detected in 110 AR patients and 101 healthy controls in Singapore Chinese population by polymerase chain reaction (PCR) and restriction fragment length polymorphism(RFLP). Results Rs729302 was rejected as it was not polymorphic. For others SNP, no statistically significant difference was detected in genotype between AR and healthy control group (X2 value were 0. 21, 5.02, 0.01, 2. 91,0. 37, all P 〉 0.05 ). No statistically significant difference was detected in allele frequencies between AR and healthy control group ( X2 value were 0. 00, 2. 78, 0. 01, 2. 31, 0.00, all P 〉 0. 05 ). Conclusion No association is observed between IRF5 and AR in Singapore Chinese population.
