摘要 目的探讨胰腺癌细胞中胶质细胞源神经营养因子(GDNF)对基质金属蛋白酶(MMP)-9表达的影响。方法用DMEM培养基进行人胰腺癌细胞株MIAPaCa-2和Capan-2细胞培养,利用RT-PCR、Westernblot和ELISA法研究胰腺癌细胞中GDNF对MMP-9mRNA及蛋白表达的影响。结果 RT-PCR实验发现胰腺癌细胞中GDNF能上调MMP-9mRNA的表达,Westernblot法、ELISA法检测结果表明胰腺癌细胞中GDNF能上调MMP-9蛋白的表达,且于GDNF浓度为100μg/L,作用48h时达最大值。结论 GDNF具有上调胰腺癌细胞MMP-9表达的作用,从而增强胰腺癌细胞的侵袭能力。 Objective To investigate the influence of glial cell line-derived neurotrophic factor (GDNF)on the expression of matrix metalloproteinase-9 (MMP-9) in pancreatic carcinoma. Methods The human pancreatic cancer cell lines MIA PaCa-2 and Capan-2 were cultured with DMEM medium. RT-PCR, Western blot and ELISA methods were used to investigate the effect of GDNF on the expression of MMP-9 mRNA and protein in pancreatic carcinoma. Results The result of RT-PCR showed that GDNF up-regulated the expression of MMP-9 mRNA. Results of Western blot and ELISA methods showed that the expression of MMP-9 mRNA was up-regulated by GDNF, and which reached the maximum value in 100 μg/L GFNF for 48 hours. Conclusion GDNF up-regulates the expression of MMP-9 and thereby strengthens the ability of invasion in pancreatic carcinoma.
