LIVIN基因调控胃癌细胞增殖及凋亡

Loss of LIVIN Expression Inhibits Proliferation and Induced Apoptosis in Human Gastric Carcinoma MKN45 Cells

目的评估LIVIN基因在不同人胃癌细胞系中的表达,并研究抑制LIVIN基因表达对胃癌细胞增殖和凋亡的影响。方法通过Western Blot检测多个胃癌细胞系中LIVIN基因的表达,筛选出高表达的细胞系。设计并合成针对LIVIN的siRNA(siLIVIN),转染胃癌细胞,分析其细胞增殖和凋亡的影响。结果 Western Blot表明MKN45细胞中LIVIN基因高表达。设计合成的siRNA转染MKN45细胞后能显著沉默LIVIN基因的表达。克隆形成实验表明siLIVIN对MKN45细胞的抑制率为63.53%(P<0.05);MTT实验表明siLIVIN对MKN45细胞的抑制率72h为47.36%(P<0.05)。TUNEL检测表明siLIVIN能引起(34.62±3.21)%(P<0.05)的细胞凋亡率。Western Blot结果表明siLIVIN能抑制抗凋亡蛋白JNK的表达和活化。结论 LIVIN在MKN45细胞中能显著促进细胞增殖,抑制细胞凋亡,LIVIN可能是一个潜在的胃癌基因治疗靶点。

Objective The goal of the present study is to analyze the expression pattern of LIVIN in a series of gastric cancer cell lines, and to estimate the potential anti proliferation and pro-apoptosis function of LIVIN in human gastric carcinoma cells. Methods Expression of LIVIN in a series of gastric cancer cell lines was estimated by Western Blot. The LIVIN targeted siRNA（siLIVIN）was designed and synthesized. The Proliferation and apoptosis of gastric cancer cell lines were measured after transfection of siI.IVIN. Results MKN45 cell line is selected with a high expression level of LIVIN. Transfection of siLIVIN significantly reduces LIVIN expression. Clone formauon assay and MTT assay show a promising inhihitory effect of siLIVIN on MKN45 cells. In addition, TUNEL assay indicated that siLIVIN expression also induced severe apoptosis. Furthermore, siLIVIN treatment also inhibits activation of pro-proliferation protein JNK. Conclusion loss of LIVIN expression markedly inhihited cell proliferation and induced apoptosis in human gastric cancer cells,and LIVIN could be a promising therapeutic target for gastric carcinoma.
