摘要 目的研究解整合素-金属蛋白酶8(ADAM8)及血管内皮生长因子受体3(VEGFR-3)在乳腺癌中的表达,探讨其与乳腺癌浸润、转移的关系,以及两者的表达特征和内在联系。方法应用免疫组织化学法检测44例乳腺癌组织及13例正常乳腺组织中ADAM8和VEGFR-3的表达,并判定ADAM8与VEGFR-3在乳腺组织中表达量。结果 VEGFR-3在乳腺癌组织中的表达明显低于正常乳腺组织(P=0.044)。肿瘤>2cm的VEGFR-3表达低于肿瘤≤2cm者(P=0.040)。Ⅲ+Ⅳ期的阳性表达低于Ⅰ+Ⅱ期(P=0.574),有淋巴结转移的乳腺癌组织ADAM8表达低于无淋巴结转移组(P=0.560),但差异无统计学意义;VEGFR-3在乳腺癌组织中的表达明显高于正常乳腺组织(P=0.000),Ⅲ+Ⅳ期的阳性表达高于Ⅰ+Ⅱ期(P=0.006),有淋巴结转移的乳腺癌组织ADAM8表达高于无淋巴结转移组(P=0.001)。肿瘤>2cm的ADAM8表达高于肿瘤≤2cm者(P=0.192),但差异无统计学意义。结论 ADAM8在乳腺癌组织中表达下调,VEGFR-3在乳腺癌中高表达,其程度与乳腺癌肿瘤分期,淋巴转移及治疗大小相关,两者可能在乳腺癌进展中起协同作用,可作为预测乳腺癌转移的指标之一。 Objective To investigate the expressions of a disintegrimlike and metalloproteinase 8(ADAM8) and vascular endothelial growth factor receptor 3(VEGFR 3)in human breast cancer and the normal breast tissue,as well as the relation of their expression pattern in tumor infiltration and metastasis. Methods The expressions of ADAM8 and VEGFR 3 were measured by immunohistochemistry in 44 breast cancer tissues and 13 normal breast tissues. Results ADAM8 staining intensity measured by IHC was significantly lower in breast cancers compared with level of normal breast tissue ( P =0. 044), and the cancerswith tumor size≤ 2 cm exhibited less VEGFR-3 expression than those with tumor size≤2 cm(P=0. 040). The VEGFR 3 expression was lower in stage III , IV as compared with those in stage I , II (P=0. 574). ,and the tissues with positive lymph node metastasis exhibited less VEGFR 3 expression than those without lymph node metastasis (P = 0. 560), although the difference did not reach statistically level. VEGFR-3 staining intensity measured was significantly higher in breast cancer compared with level of normal breast tissue(P= 0. 044) ,The VEGFR 3 expression was higher in stage III ,IV as compared with those in stage I , II (P=0. 006) ,and the tissues with positive lymph node metastasis exhibited higher VEGFR 3 expression than those without lymph node metastasis(P=0. 001 ). The tissues in tumor size≤2 cm exhibited more VEGFR-3 expression than those in tumor size≤2 cm(P=0. 192). Conclusions The low expressions of ADAM8 and the high expressions of VEGFR 3 were observed in breast cancer tissues,and their staining intensity was positively related to tumor stages,tumor lymph node metastasis and tumor size. They appear to work synergistically in the progression of breast cancer,and implied in in the prognosis of this disease.
