摘要
目的研究肝硬化大鼠模型肠黏膜相关淋巴组织T淋巴细胞及其亚群的表达,了解肝硬化肠黏膜免疫屏障功能的变化。方法采用皮下注射40%四氯化碳建立大鼠肝硬化模型,分离肠上皮内、固有层、派伊尔结和肠系膜淋巴结淋巴细胞,分别标记小鼠抗大鼠CD3、CD4、CD8单克隆抗体,流式细胞仪检测T淋巴细胞及其亚群的表达情况。结果与正常组相比,肝硬化组肠上皮内CD3+T淋巴细胞比例有所减少,但无统计学差异(P>0.05);CD4+CD8-亚群细胞比例减少(P<0.05),CD4+CD8+亚群细胞比例增加(P<0.05);肝硬化组固有层、派伊尔结、肠系膜淋巴结CD3+T淋巴细胞比例均明显降低(P<0.05或P<0.01)。结论肝硬化时肠黏膜相关淋巴组织细胞免疫功能降低。
Objective To study the expression of T lymphocyte and its phenotypes in the gut - associated lymphoid tissues of the cirrhotic rat models, and to understand the changes of the function of intestinal mucosa immunologic barrier. Methods The rat model of hepatic cirrhosis was made through the subcutaneous injection of 40% carbon tetrachloride. The lymphocytes of the intestinal epithelium,lamina propria, Peyerg patches, and mesenteric lymph node were isolated and labeled with mouse anti - rat CD3, CIM, and CD8 monoclonal antibody. Flow cytometry was used to detect the expression of the T lymphocyte and its phenotypes. Results Compared with the proportions of the control group, the proportion of intestinal intraepithelial CD3 + T lymphocyte in the cirrhotic group decreased, but the difference was not significant( P 〉 0.05 ) ; the proportion of CD4 + CD8- subset ceils decreased (P 〈 0.05 ), while the proportion of CD4 + CD8 + subset cells increased ( P 〈 0.05 ). In the cirrhotic group, the proportions of CD3 + T lymphocytes in the lamina propria, Peyer's patches, and mesenteric lymph node all significantly decreased ( P 〈 0.05 or P 〈 0.01 ). Conclusion The cellular immune function of gut - associated lymphoid tissues decreases along with the occurrence of hepatic cirrhosis.
