子痫前期患者外周血中Th17、Treg细胞变化及意义 被引量:12

Changes of Treg and Th17 Cells in Peripheral Blood in Preeclampsia Patients and its Signification

摘要 目的研究CD4+CD25+Foxp3+调节性T细胞(CD4+CD25+Foxp3+Treg)以及Th17细胞在子痫前期(PE)患者外周血比例的变化,并探讨其在子痫前期免疫耐受失衡中的作用。方法选取子痫前期患者20例,同期正常晚期妊娠20例。分别采集子痫前期患者、正常晚期妊娠妇女外周血,采用细胞内染色的流式细胞术检测CD4+CD25+Foxp3+Treg以及CD4+IL-17A+的表达。结果子痫前期患者外周血CD4+CD25+Foxp3+/CD4+细胞的比例明显低于正常晚期妊娠者(P<0.05);而CD4+IL-17A+/CD4+细胞的比例明显偏高(P<0.05)。结论子痫前期患者上调Th17的表达,Treg细胞表达下降,可能产生Th17/Treg平衡偏移,这一结果造成母胎免疫耐受失衡,导致子痫前期的发生。 Objective To observe the changes of CD4+CD25+Foxp3+Treg and Th17 cells in peripheral blood in preeclampsia (PE) patients , and explore its immuneunbalance function in PE. Methods Peripheral blood of 20 cases with PE and 20 cases of normal gravida with late pregnancy were collected. The level of CD4+CD25+Foxp3+Treg and CD4+IL- 17A+ was detected by intracellular staining flow cytometry. Results CD4+CD25+Foxp3+Treg in CD4q-cells percentages in peripheral blood of PE patients were significantly lower than that in normal gravida with late pregnancy. While,CD4+ IL- 17A+ in CD4+cells percentages in peripheral blood of PE patients were higher. Conclusion The expression of Th17 cells was higher,Treg cells was lower in PE patients. The balanced of Th17 and Treg may migrate. It indicates that maternal fetal immune tolerance becomes imbalance,leading to the occurrence of preeclampsia.
