摘要
以省沽油叶为原料,采用酸水解、乙醇沉淀法对省沽油叶中的复合氨基酸提取工艺进行研究,并用电位滴定法对所得产物进行含量测定。结果表明:省沽油叶子中的复合氨基酸提取以液固比(mL/g)2∶1、盐酸体积分数25%、水解时间10 h、水解温度110℃效果为好;脱色工艺以活性炭加入量占省沽油叶量的12%,并在(75±5)℃的温度下脱色40 min效果为好;当pH值为3,无水乙醇用量为待分离液体积的1.25倍时进行无水乙醇沉淀,所得复合氨基酸产物的AN含量最高。
In this study,acid hydrolysis and ethanol precipitation were applied to extract compound amino acid from bumalda bladdernut(Staphylea bumalda DC) leaves.The nitrogen content of the extracted product was determined by means of potentiometric titration.The results showed that the optimum extraction parameters were liquid-to-solid ratio of 2∶1,hydrochloric acid concentration of 25%,hydrolysis time of 10h and temperature of 110℃.The optimal decoloration effect was achieved when it was conducted at(75±5)℃ for 40 min with activated carbon weighted 12% of bumalda bladdernut leaf weight.The highest amino nitrogen(AN) content of compound amino acid was obtained when the extraction was carried out in acid condition with a pH value of 3 and ethanol was 1.25 times the volume of the treated liquid.
