The Relationship of Centrosome Amplification in Oral Squamous Cell Carcinoma to the Expression of CyclinE and p21^(waf1) Proteins

摘要 目的:了解口腔鳞状细胞癌(OSCC)cyclinE及p21waf1蛋白表达与中心体扩增相关性,探讨OSCC中心体扩增的分子机制。方法:8例正常口腔黏膜及27例OSCC石蜡包埋组织,采用流式细胞术对cyclinE和p21waf1蛋白表达进行定量检测,并分析其与相应组织中心体扩增的相关性。结果:OSCC中心体扩增与cyclinE/p21waf1比值呈正相关(r=0.679,P<0.01),而与p21waf1蛋白表达量呈负相关(r=-0.472,P<0.05)。结论:cy-clinE/p21waf1比值增高可能是OSCC中心体扩增的原因之一,p21waf1蛋白表达下调在其中可能起主要作用。 Objective: To understand the relationship of cyclinE and p21^wafl proteins to centrosome amplification in oral squamous cell carcinoma (OSCC), and so as to explore the possible molecular mechanism of centrosome amplification in OSCC. Methods: The expression levels of cyclinE and p21^wafl proteins in paraffin-embedded tissues of 8 cases normal oral mucosa and 27 cases with OSCC were quantitatively detected with flowcytometry. The correlation between the expression of these two proteins and centrosome amplification in OSCC was statistically analyzed. Results: Centrosome ampli- fication was positively correlated with the ratio of cyclinE/p21^wafl ( r = 0. 679 ,P 〈 0.01 ), but was nega- tively correlated with the expression level of p21^wafl protein ( r = - 0. 472, P 〈 0.05 ). Conclusions: Up-regulated ratio of cyclin E/p21^wafl proteins may play an important role in centrosome amplification in OSCC and down-regulated p21^wafl protein level may be a major factor in it.
