摘要
目的观察孤儿核受体神经生长诱导基因B-β(nerve growth factor-induced gene B-β,NGFIB-β)在大鼠大脑皮层神经细胞分化中表达的动态变化,探讨NGFIB-β在神经细胞分化成熟中的表达意义。方法选取成熟大鼠5只,孕12~19d妊娠大鼠(每天5只)、生后1~15d幼鼠(每天5只),处死后取大脑皮层组织,采用石蜡包埋组织切片,免疫组织化学染色测定NGFIB-β表达变化。结果 NGFIB-β从孕16天开始在大鼠大脑皮层第Ⅴ、Ⅵ层出现少量阳性细胞,随着发育的进行阳性细胞数目明显增多,生后天阳性表达达到高峰,在Ⅱ-Ⅵ均见阳性细胞,之后随着细胞的分化成熟阳性细胞数目又明显减少,成熟的大脑皮层仅在Ⅴ-Ⅵ层见少量阳性细胞。结论提示NGFIB-β可能在大鼠大脑皮层神经细胞从幼稚到成熟的分化过程中有调控作用。
Objective To observe the dynamic changes of the expression of nerve growth factor-induced gene B-β(NGFIB-β)in nerve cell differentiation of cerebral cortex of rats,and to investigate the significance of expression of NGFIB-β in the differentiation, migration and maturation of nerve cells. Methods The 15 mature rats,40 pregnant rats during 12 - 19d of pregnancy,with 5 rats in each day, the 75 young rats during 1 - 15d after birth, with 5 rats in each day, were enrolled in the study. The rats were sacrificed respectively, then the rats' cerebral cortex tissues were taken out and prepared into paraffin-embedded sections, the expression of NGFIB-β was detected by immunohistochemistry. Results A few NGFIB-βpositive ceils were found at layers Ⅴ, Ⅵ of cerebral cortex from 16d after pregnancy, the positive cells were obviously increased with the development, which reached peak at 1,2,3,4,5d after birth. The NGFIB-βpositive cells were observed at layers Ⅱ ,Ⅵ , later the positive cells were obviously decreased with the differentiation and mature, and only a few positive cells could be found at layers Ⅴ, Ⅵ in mature cerebral cortex. Conclusion NGFIB-β may have a regulatory role during the differentiation, migration and maturation of cerebral cortex nerve ceils in rats.
