摘要
目的研究前列腺癌组织中缺氧诱导因子-1α(Hypoxia inducible factor-1α,HIF-1α)、微血管密度(Microvessel density,MVD)的表达情况,探讨HIF-1α、MVD在前列腺肿瘤发展中的作用。方法对72例前列腺癌组织标本采用免疫组化SP法,分别检测不同病理级别前列腺癌组织中HIF-1α表达情况与MVD计数情况。结果 72例前列腺癌组织中HIF-1α的表达阳性率为69.4%(50/72)。在高分化、中分化和低分化的前列腺癌组织中HIF-1α的阳性表达率差异有统计学意义(χ2=52.06,P<0.01)。HIF-1α的表达强度和前列腺癌的病理分级有相关性(r=0.49,P<0.05),分化程度越低,HIF-1α表达越高。MVD计数在不同病理分级前列腺癌组织之间差异有统计学意义(F=282.45,P<0.01)。结论前列腺癌组织中HIF-1α的表达和MVD计数均与病理级别密切相关,提示HIF-1α的过度表达和MVD的增高可能在前列腺癌的发生、发展、侵袭、转移等生物行为过程中起重要作用,为以HIF-1α为靶点治疗前列腺癌提供理论依据。
Objective To investigate the expression of hypoxia inducible factor-1α (HIF-1α )and micro-vascular density (MVD)in prostate cancer, and to explore their roles in the pathogenesis and development of prostate cancer. Methods The expression of HIF-1α and MVD counting were detected by immunohistochemistry (SP) in 72 specimens of prostate cancer tissues which were different pathological grades. Results The positive expression rate of HIF-1α in 72 specimens of prostate cancer tissues was 69.4% ( 50/72 ). There was a significant difference in the positive expression rate of HIF-1α among different ( high, medium, low) differentiation groups which were 95.83% , 64. 29% , 45.00% , respectively ( P 〈 0. 01 ). The expression intensity of HIF-1α was negatively correlated with pathology grade of prostate cancer ( r = 0.49, P 〈 0. 05 ). There was a significant difference in MVD counting among different (high, medium, low)pathological grading groups which were 11.40 ± 2.70,29.00 ±1.87,49.20 ± 2.86, respectively ( F = 282.45, P 〈 0.01 ). Conclusion The expression of HIF-1α and MVD counting in prostate cancer tissues are closely related with different pathological grades, which suggests that overexpression of HIF-1α and increase of MVD may play an important role in the pathogenesis, progression, invasion, metastasis of prostate cancer, and the experimental results provide theoretical basis for the treatment of prostate cancer by taking HIF-1α as target.
