摘要
以‘粉娇蓉’(Lagerstroemia indica‘Fen Jiaorong’)、‘红红火火’(L.indica‘Honghong Huohuo’)、‘堇薇’(L.indica‘Jinwei’)、‘直枝紫’(L.indica‘Zhizhi Zi’)4个常见紫薇(L.indica)品种的混合花粉为材料,采用正交试验对影响紫薇花粉生活力的3个因素(蔗糖、硼酸和CaCl2)进行优化试验,依据正交试验不同组合的花粉萌发率,确定最佳花粉萌发组合.在此基础上,对18种不同贮藏条件下花粉耐贮藏性进行检验.结果表明,用200 g.L-1蔗糖+150 mg.L-1硼酸+20 mg.L-1CaCl2为最佳培养基,3个因素对紫薇花粉萌发率的影响从大到小顺序为蔗糖、硼酸、CaCl2;不同贮藏处理间花粉耐贮藏力存在较大差异,18个处理组合中,室温(28℃)条件下,用变色硅胶干燥的处理生活力保持时间最长;4℃冷藏条件下的对照、遮光、干燥处理保持时间稍短,但贮藏期间能保持较高的萌发率.
Using the mixed pollen of four common breeds Lagerstroemia indica, L. indica 'Fen Jiaorong' , L. indica 'Honghong Huohuo' , L. indica ' Jinwei' and L. indica 'Zhizhi Zi' as materi- als, and adopting the orthogonal test methods optimizing experments were conducted on the three factors affecting pollen viability (sucrose, boric acid, and CaCl2) to determine the best combination of pollen germination according to the result of orthogonal experiment. And on that basis, pollen storage toler- ance was tested under 18 storage conditions. The results were as follows: the optimal culture medium was 200 g . L-1 sucrose + 150 mg . L-1 boric +20 mg . L-1 CaCl2 ;sucrose showed the most obvious effect on pollen germination rate of Lagerstroemia indica followed by boric and CaCl2 ; there are differ- ences in pollen storage tolerance with different storage conditions. During the 18 kinds of methods, the one dealing with allochroic silica gel under the normal temperature of 28℃ could keep the longest pol- len viability; another one dealing with CK, shading, drying under the refrigerated condition of 4 ℃would keep a shorter time , but could get a higher pollen germination rate during storage.
