摘要
用抗癌免疫核糖核酸(iRNA)在人体内诱生IFN—α,并经提纯获得高活性IFN—α,用于癌细胞的抑制试验,并将IFN-α标记为^(125)I—IFN—α,检测癌细胞表面IFN—α受体。结果表明,提纯的IFN—α对胃癌7901细胞、Hep—2细胞、Hela细胞、L929等细胞于37℃4h孵育后,细胞膜上有^(125)I—IFN—α吸附,并与IFN—α有竞争性抑制,表现在cpm数值的增高或降低。
The IFN-α with high activity induced in human by anti - cancerous iRNA was used in cancer cell inhibitory test and labelled as 125I -IFN-α to detect the IFN -α recepor on the cancer cell surface. The results showed that 125I-IFN -α can be absorbed to the membrane of gastric cancer 7901 cells, Hep-2 cells, Hela cells and L929 cells after incubation at 37℃ for 4h had a competitive inhibitory effect on cancer cells expressed as the increase and decrease of cpm values.
