抗腺病毒单克隆抗体免疫酶染色特性的研究

STUDY ON THE CHARACTERISTICS OF IMMUNOENZYMATIC STAINING OF MONOCLONAL ANTIBODIES AGAINST ADENOVIRUS

对六个抗腺病毒型特性单克隆中和抗体(MeAb)(7-1-1、7-1-6、7-4-1、7-4-4、3-4-2、3-9-1)的免疫酶染色特性进行了系统的研究,并与常规抗体进行了比较。这些McAb,不引起非特异性染色,对腺病毒抗原的染色特异性几乎是绝对的,但型特异性是相对的,有着浓度依赖性;亲和力高的McAb的染色强度可接近常规抗体,并出现“前带现象”;两个针对不同抗原决定簇的McAb_s组成的“合剂”,在本实验的具体条件下没能增加染色强度,对此进行了讨论。

The characteristics of immunoenzymatic staining of six types of specific monclonal antibodies (McAbs) against adenovirus type 3 or 7 were compared with those of the conventional antibodies and systematically studied. 1. No non-specific stainig was observed in these McAbs. The specificity of the McAbs to antigen of andenovirus is almost absolute, but their type-specificity is relative, dependent upon the concentration of McAbs. The type-specificity will be lost and cross-reaction between different types of adenovirus may occur if the concentration of the McAbs is very high. 2. The staining intensity of high affinity McAbs is close to that of the conventional antibodies. The 'pro-zone ' phenomenon may also occur when using these McAbs for staining. The staining intensity was not increased when two different epitypes neutralizing McAbs were used in combination.
