摘要
现介绍5种常用的单克隆抗体(McAb)制备技术,即杂交瘤技术、嵌合抗体制备技术、噬菌体抗体库技术、转基因小鼠制备抗体技术和核糖体展示技术,并对这5种技术的优缺点进行分析。阐述并比较了McAb制备所依赖的表达系统,其中包括细菌表达系统、酵母和丝状真菌表达系统、昆虫杆状病毒表达系统、植物表达系统和哺乳动物表达系统。最后,展望了McAb未来的发展前景及其在生物技术和疾病诊断方面上的应用。
Five kinds of commonly used monoclonal antibody producing technology, namely the hybrido- ma technique, the chimeric antibody producing technology, phage antibody library technology, transgenic mice antibody producing technology and ribosome display technology are introduced, and their merits are analyzed. Then, five monoclonal antibody-dependent expression systems are illustrated and compared, including bacteri- al expression system,yeast and filamentous fungi expression systems, baculovirus expression system, plant ex- pression system and mammalian expression system. Finally, the future of the monoclonal antibody develop- ment and its application in the biotechnology and disease diagnosis are prospected.
