耐寒茵陈蒿试管苗培养的研究 被引量:1

摘要 为保存和利用耐寒茵陈蒿资源,以具有侧芽或顶芽的分枝嫩茎为材料,采用组织培养的方法,对茵陈蒿进行了生根培养、生根继代增殖培养以及试管苗的移栽和定植的研究。结果表明:1/2MS+NAA 0.08mg.L-1是具有顶芽或侧芽分枝嫩茎段生根培养生长为试管苗的适宜培养基;向培养瓶中加入10mL浓度为15mg.L-1的NAA溶液处理4h后,把具有2个侧芽或者具有1个顶芽和1个侧芽的试管苗茎段,接种到1/2MS培养基上进行生根增殖培养的方法是试管苗生根增殖培养的理想方法。试管苗移栽成活率在96.0%以上;定植成活率在99%左右;定植的试管苗不仅保持了野生茵陈蒿的所有植物学性状,而且还保持了耐寒变异性状。 To preserve and use frigostabile Arternisia capillaris resources, with the method of tissue culture, tender stems with lateral bud and terminal bud of Artemisia capillaris were used as material to conduct the re search on the rooting culture, root successive, transplantation of tube seedlings and stable planting of tube seed lings. The results showed that the ideal medium for rooting from lateral bud and terminal bud to tube seedlings was 1/2MS+NAA 0.08 mgoL1. The ideal method was that inoculated two lateral buds or one lateral bud and one terminal bud which had been dealt with 10 mL 15 mg.U1 NAA for 4 hours into 1/2MS. The percentage of transplant survival had been over 96.0% and the percentage of stable planting of tube seedlings had been al most 99. 0%. The tube seedlings stable planted maintained all biological characteristics of the wild and the vari ation character of frigostabile.
