摘要
枯草芽孢杆菌CG24是一株烟草野火病菌拮抗菌,本文采用单因素实验和响应面法对该菌株发酵条件进行了优化。结果表明CG24菌株在培养温度32℃初始pH 7.5装液量50 mL/150 mL和接种量4%的NA培养液中振荡培养28 h,其代谢产物的抑菌活性作用最为显著。通过单因素实验确定了影响菌株CG24生长的培养基主要因素为葡萄糖、酵母膏和氯化钙,对3个因素进行响应面优化分析,确定最优发酵培养基为:葡萄糖40 g/L酵母膏7.55 g/L氯化钙0.72 g/L,抑菌圈直径比优化前提高了25.72%,为该菌株以后的扩大培养和微生态制剂的开发应用奠定了基础。
Bacillus subtilis s an antagonistic bacterium to tobacco wild fire. The fermentation condition subtilis CG24 optimized through single factor test and response surface method. The results showed that the metabolites of antibacterial activity was pronounced in the culture of 32℃, the initial pH 7. 5, liquid volume of 50 mL/150 mL and 4% inoculation quantity cultured 28 h. By the single factor experiment, the main factors f strain CG24 were the glucose, yeast extract and calcium chloride. By response surface optimization analysis, the optimal fermentation medium was glucose 40 g/L, yeast extract 7.55 g/L, calcium chloride 0. 72 g/L. The optimization of formulation antibacterial circle diameter was improved by 25.72%. It lays a foundation for the strain incubation and micro-ecological preparation application.
