Distribution of Sliver in Plasma and Tissues of Burnt Rats with Nano-silver Cream

摘要 目的:研究纳米银乳膏在烧伤模型大鼠血浆及组织中银的分布。方法:建立大鼠5%体表总面积(5cm×5cm)深Ⅱ度蒸气烧伤模型,分为纳米银乳膏组、磺胺嘧啶银乳膏组、基质对照组,每组8只,分别于受伤感染处涂药0.1~0.2cm厚,每日1次,连续30d,停药24h后处死大鼠,利用电感耦合等离子体质谱仪测定各组大鼠血浆和心、肝、脾、肾组织中的银含量。仪器条件:射频功率为1250W,脉冲电压为1200V,雾化气流速为0.80L/min,等离子气流速为15L/min。结果:银检测质量浓度的线性范围为0.01~10.00μg/m(lr=0.9999),加样回收率为(93.2±3.3)%~(112.5±1.0)%,RSD<3.5%(n=8)。与基质对照组比较,磺胺嘧啶银乳膏组大鼠的血浆和心、肝、脾、肾组织中银含量均明显增加(P<0.05或P<0.01),而纳米银乳膏组大鼠仅脾组织中银含量明显增加(P<0.05)。结论:纳米银乳膏在大鼠血浆和除脾以外的各组织中无明显蓄积。 OBJECTIVE: To study the distribution of sliver in plasma and tissues of burnt rats with Nano-silver cream. METH- ODS: Deep Ⅱ degree scald model of 5% total body surface area (5 cm×5 cm) of rats was established and randomly divided into Nano-silver cream group, Flamazine cream group and base control group with 8 mrs in each group. They were applied cream to in- jured infection site coating with 0.1-0.2 cm, once a day for consecutive 30 d. Rats were sacrificed 24 h after drug withdrawal. The contents of sliver in plasma, heart, liver, spleen and kidney were determined by ICP-MS. The condition of determination was as follows: radio-frequency power 1 250 W, impulse voltage 1 200 V, flow velocity of atomizing gas 0.80 L/min, flow velocity of plasma gas 15 L/min. RESULTS: The linear range of sliver was 0.01-10.00μg/ml (r=0.999 9) with recoveries of (93.2 ±3.3)%- (112.5 ± 1.0)% (RSD〈3.5%, n=8). Compared with base control group, the contents of sliver in plasma, heart, liver, spleen and kidney increased significantly in Flamazine cream group (P〈0.05 or P〈0.01), while the contents of sliver in only spleen in- creased significantly in Nano-silver cream group (P〈0.05). CONCLUSIONS: Results show that no significant accumulations of Na- no-silver cream in plasma and tissue except spleen of rats.
