人乳头瘤状病毒P53基因与膀胱癌关系的研究 被引量:2

摘要 目的HPV与膀胱癌的研究存在很多争论,尤其是HPV在膀胱癌组织中的检出率有很大差异,O%到81%不等,本课题旨在明确HPV、P53与膀胱移行细胞癌发生的关系。方法PCR体外扩增和DNA反向点杂交相结合的DNA芯片技术测定39例膀胱移行细胞癌中高危型HPVl6、18、31、33、35、39、45、51、52、53、56、58、59、66、68、73、83、MM4;低危型HPV6、11、42、43、44-等亚型的感染情况,同时用免疫组化测定P53的表达情况。结果膀胱癌组织的HPV总体阳性率为23.1%,HPV的亚型主要为11,16,其中有3例为HPVll和HPVl6的混合感染,有一例检测到HPVl8的亚型18,HPVl6最常见,占66.7%。HPV的感染与P53表达呈正相关,与肿瘤分级无关。结论膀胱移行细胞癌发生与HPV感染的确存在明显的相关性,其中HPV亚型以HPVl6为最常见,HPVll也是诱发肿瘤的亚型之一,HPV诱发肿瘤的原因可能是干扰抑癌基因P53功能,从而诱发肿瘤发生。 Objectives There are many issues in the relationship between transional cell carcinoma and HPV as well as P53 , especially, The prevalence of HPV in TCC is still inconsistent,so,It is important to study defi- nitely the relationship between transional cell carcinoma and HPV as well as P53. Methods HPVs in 39 Cases of transional cell carcinoma were analyzed by PCR and Reverse hybridization in situ. The HPVs included high risk types HPV16 ,18 ,31,33 ,35 ,39 ,45 ,51,52 ,53 ,56 ,58 ,59 ,66 ,68 ,73 ,83 , MM4 ; low risk types : HPV6 ,11,42 ,43 ,44 , at that time, P53 gene in these tissue were detected by immunochistochemieal method. Results Total Positive rate of HPV in TCC of bladder was 23.1% ,HPV16 and HPVll were major types,HPVll and HPV16 were both found out in 3 cases . HPV16 is the best common type, Its proportion is 66.7%. There was a positive relativity between HPV and P53, but the relativity without grade of TCC. Conclusions HPV16 and HPVI 1 can be a factor leading to occur TCC of bladder, It could be reason that Hives interfere in function of P53.
