骨折愈合过程中骨形态发生蛋白与细胞外基质磷酸糖蛋白表达的相关性 被引量:6

摘要 目的探讨骨折愈合过程中骨形态发生蛋白(BMP-2)与细胞外基质磷酸糖尿蛋白(MEPE)表达的相关。方法建立兔股骨骨折模型,通过RT-PCR检测骨折愈合过程不同阶段BMP-2与MEPE基因表达水平。随后将含有BMP-2基因的质粒转染MC3T3-E1细胞,采用免疫荧光染色方法验证BMP-2与MEPE的关系。结果骨折愈合第6天可检测到BMP-2 mRNA表达量明显增加,MEPE mRNA表达随BMP-2增加而增加;24 d后,两者下降至最初水平。体外实验结果显示,转染BMP-2的细胞MEPE表达也随之增加。结论 BMP-2在骨折愈合早期发挥重要作用,BMP-2可促进MEPE表达,是MEPE上游调节因子。
