褪黑素对结肠癌细胞RKO增殖和迁移的影响及其可能的分子机制

摘要 目的探讨褪黑素(Mel)对RKO细胞凋亡和迁移的影响及其过程中可能的分子机制。方法 MTT法检测Mel对RKO细胞增殖的影响,Western blot检测内质网应激(ERS)相关蛋白的表达水平,通过划痕实验检测Mel对RKO细胞迁移的影响,并检测骨桥蛋白(OPN)的表达情况。结果在Mel作用下,RKO细胞的增殖受到抑制,同时重链结合蛋白(BIP)的表达下降、C/EBP同源蛋白(CHOP)和caspase 3的表达水平上升;Mel抑制RKO细胞迁移,并且抑制OPN的表达。结论 Mel通过ERS途径促进RKO细胞凋亡,可能通过降低OPN蛋白的表达抑制细胞迁移。 Objective To study the effects of melatonin (Mel) on the proliferation and migration of colorectal cancer cell line RKO and the probable molecular mechanisms. Methods MTT assay was performed to measure the proliferation of RKO. Western blot was done to detect the proteins associated with the ER stress(ERS) and apoptosis. The effect of Mel on the migration of RKO cells was analyzed by cell migration test and the expression of osteopontin(OPN) was detected by Western blot. Results Mel inhibited the proliferation and promoted the apoptosis in RKO. BIP was up-regulated while CHOP and caspase 3 were down-regulated in RKO cells treated with Mel. Mel also inhibited the cell migration and the expression of OPN. Conclusion Mel promotes the apoptosis of RKO cells by ERS and inhibits the migration of RKO cells, which may be related to the down - regulation of the expression of OPN protein.
