hTERTC27过表达对鼻咽癌C666-1细胞株增殖和凋亡的影响 被引量:2

摘要 目的研究外源表达hTERTC27对鼻咽癌C666-1细胞增殖和凋亡的影响。方法采用脂质体将pcDNA3-hTERTC27转染至C666-1细胞株中。采用Western blot法鉴定hTERTC27和Bcl-2在细胞中的表达。采用MTT比色法和细胞克隆形成实验观察其对细胞增殖的影响。采用TUNEL实验观察其对细胞凋亡的影响。结果脂质体转染24h后,C666-1细胞株外源性hTERTC27出现高表达。过量表达hTERTC27的C666细胞增殖能力明显减缓(P<0.01),克隆形成率降低(P<0.01),TUNEL阳性细胞率增加(P<0.01),Bcl-2表达量降低(P<0.01)。结论 hTERTC27可以抑制C666-1细胞增殖、促进凋亡,可能与下调Bcl-2蛋白的表达有关。 Objective To explore the effect of hTERTC27 overexpression on the proliferation and apopto- sis of nasopharyngeal carcinoma C666 1 cell line. Methods pcDNA3-hTERTC27 was transfected into na sopharyngea[ carcinoma C666-I cells with lipofectamine. The expression of hTERTC27 and Bcl 2 were detected by Western blot. The proliferation of cells was detected by MTT assay and colony formation as- say. The apoptosis of cells was detected by TUNEL assay. Results C666 1 ceils expressed high level of hTERTC27 after 24h transfection. Over expression of hTERTC27 obviously suppressed the cell prolifera- tion( P〈0.01),downregulated the colony formation rate(P〈0.01 ),increased the TUNEL positive cells (P〈0.01) ,and downregulated the expression level of Bcl-2 protein(P〈0. 01). Conclusion Overexpres sion of hTERTC27 can effectively inhibit proliferation and promote apoptosis of nasopharyngeal carcinoma cells,which is possibly due to the down regulation of Bcl -2 protein.
