大熊猫源枝孢样枝孢霉的生物学特性 被引量:6

Biological characterisation of Cladosporium cladosporioidesisolated from giant panda

摘要 将大熊猫源枝孢样枝孢霉在不同培养基以及不同温度、不同pH、不同碳源和不同氮源的培养基中培养,以十字交叉法研究了各因子对其生长的影响。结果表明,该菌的最适培养基是马铃薯葡萄糖琼脂培养基和沙氏琼脂培养基,较差的是改良大米琼脂培养基和玉米琼脂培养基,最差的为子囊孢子培养基和察氏琼脂培养基。该菌最适生长温度是25℃,较差的是15℃和20℃,30℃和35℃中几乎不能生长。最适pH为pH5和pH6,pH4和pH8~pH11中生长稍差,最差的是pH7。不同碳源对该菌生长无显著差异。最适合该菌生长的氮源为硝酸钠,其次是草酸,生长较差的是硫酸铵,在尿素中不能生长。结果明确了枝孢样枝孢霉生长的最适宜环境,可为该菌引起的暗色丝孢霉病的诊断及防治提供参考。 Cladosporium cladosporioides isolated from giant panda was incubated in different media,at different temperatures or pH,in czapek dox agar(CZA) media with different carbon sources or nitrogen sources to study the influence factors on C.cladosporioides growth by using cross method.The optimal media for C.cladosporioides were potato dextrose agar and sabouraud dextrose agar,poor ones were modified rice grain agar and corn meat agar.Ascospore agar and CZA were not suitable for C.cladosporioides.The optimal temperature for C.cladosporioides growth was 25 ℃,poor ones were 15 ℃ and 20 ℃,and it could not grow at 30 ℃ and 35 ℃.The optimal pH values for C.cladosporioides growth were pH5 and pH6,poor ones were pH4 and pH8 to pH11,and the worst was pH7.There was no significant difference in the CZA media with different carbon sources.The optimal nitrogen source for C.cladosporioides growth was sodium nitrate.C.cladosporioides growing in oxalate and ammonium sulfate was bad.And it could not grow in carbamide.The present study identified the optimal growth in vitro factors for C.cladosporioides,and provided references for prophylaxis and diagnosis of phaeohyphomycosis.
