LncRNA在缺氧诱导的胃癌细胞中表达谱的变化 被引量:8

摘要 目的:观察长链非编码RNA(lncRNA)在常氧条件下的胃癌细胞及缺氧诱导的胃癌细胞中表达谱的变化。方法:利用lncRNA芯片技术检测常氧条件下的胃癌细胞,缺氧诱导的胃癌细胞中lncRNA表达谱的变异,经对原始数据进行预处理达到均一化后,筛选出差异表达lncRNA,进行分析。结果:与常氧条件下的胃癌细胞相比较,缺氧诱导胃癌细胞中1.5倍以上变化并有显著差异(P<0.05)的lncRNA,确定为差异表达lncRNA。其中在三种胃癌细胞中的变化均在1.5倍以上的共254条,占所有lncRNA的3.6%;大于1.5倍上调的共136条,1.5倍以上降低的共118条;2倍以上上调的共30条;2倍以上降低的共25条;3倍以上升高的共5条;3倍以上降低的共5条。结论:缺氧诱导的胃癌细胞与常氧的胃癌细胞比较,lncRNA表达谱发生显著变化。提示差异性表达的lncRNAs可能参与了缺氧环境下胃癌细胞多种恶性表型的变化。 Objective :To analyze the expression profile variation of long non -coding RNA (lncRNA) in gastric cancer cell lines under normoxia and hypoxia. Methods: To use lncRNA microarray technology to inspect the difference of lncRNA expression profile between the gastric cancer cell lines under normoxia and hypoxia, screening out the lneRNA with diferential expression after pretreatment and homogenization of raw data and make hierarchical clustering and volcano scatter diagram analysis. Results: Compare with the gastric cancer cell lines under normoxia , and 254 lncRNA which have more than 1.5 times variation and significant diference ( P 〈 0.05 ) by statistical analysis are regarded as lncRNA with differential expression, accounting for 3.6% of all lncRNA:while 136 increase more than 1.5 times and 118 reduce more than 1.5 times;30 increase more than 2 times:25 reduce more than 2 times;5 increase more than 3 times;and 5 reduce more than 3 times. Conclusion: LncRNA expression profile of the gastric cancer cell lines under hypoxia changes significantly in comparison with the gastric cancer cell lines under noemoxia. The different lneRNAs may include a fair proportion of lneRNA exerts enhancer - like effect and lneRNA exerts inhibitory effect, participating in the critical section of regulatory mechanism in the gastric cancer cells under hypoxia.
