微量元素肥料和茉莉酸甲酯处理对金银花有效成分含量的影响 被引量:8

Influences of micronutrient fertilizer and methyl jasmonate treatments on the content of compounds in Lonicera japonica

摘要 目的:应用高效液相色谱法(HPLC),研究不同微量元素肥料(微肥)、不同浓度茉莉酸甲酯(MeJA)处理对金银花中7种有效成分含量的影响。方法:应用转换波长的高效液相色谱分析方法,色谱柱为Agilent Eclipse Plus C18柱(250mm×4.6mm,5μm);流动相为0.3%甲酸水溶液(A)和乙腈(B),进行梯度洗脱;流速1mL/min;采用VWD紫外检测器转换波长(330、350nm)检测;柱温为常温。结果:不同微肥、不同浓度MeJA处理能不同程度地影响金银花中各个成分或总成分含量。结论:不同微肥处理中,Fe处理可同时增加绿原酸、木犀草苷含量;不同浓度MeJA处理中,浓度为800、1 600μmol/L的MeJA可增加绿原酸及总成分含量。 Objective: A high-performance liquid chromatographic(HPLC) method was developed to research the influences of micro-elements and methyl jasmonate treatments at the content of 7 kinds of compounds in Lonicera japonica. Methods: HPLC was developed to determine the contents of seven compounds in Lonicera japonica and folium lonicerae with wavelength conversion, analysed the content differences between them. Chromatographic column was a C18(250mm × 4.6mm, 5 μ m) column operating at normal temperature with a flow rate of lmL/min, and detection wavelength at 330, 350nm. The mobile phase using gradient flow consisted of two solvent systems: 0.3% methanoic acid and acetonitrile. Result: Different kinds of micro-elements and different concentration of methyl jasmonate had an certain impact on each or total content of compound in Lonicera japonica. Conclusion: In treatments of different kinds of micro-elements, Fe treatment can simultaneously increase the contents of chlorogcnic acid and luteoloside; in treatments of different concentrations of methyl jasmonate, concentration of 800μmol/L, 1 600μmol/L can increase the contents of chlorogcnic acid and total contents.
