β_3肾上腺素能受体基因多态性与2型糖尿病、原发性高血压发病相关性的研究 被引量:5

摘要 目的研究β3肾上腺素能受体(β3-AR)基因第一外显子第190位点T/C多态性与2型糖尿发病、原发性高血压发病的相关性。方法采用病例对照研究的方法,以74名2型糖尿病患者、82名原发性高血压患者与108名健康中老年人的空腹肘静脉血样本,分离白细胞并提取DNA,应用聚合酶链反应(PCR)-限制性酶切片段长度多态性技术检测两组的β3-AR基因第一外显子第190位点T/C多态性,比较三组各自的基因型和等位基因的分布频率。结果β3-AR基因第一外显子第190位点T/T、T/C和C/C基因型在2型糖尿病组中分别为68.92%、17.57%、13.51%,在原发性高血压组中分别为82.93%、13.41%、3.66%,在正常中老年人组中分别为84.26%、12.96%、2.78%。2型糖尿病组与原发性高血压组及正常中老年人组的基因分布频率差异均有显著性,而原发性高血压组与正常中老年人组的基因分布频率差异无显著性。结论β3-AR基因第一外显子第190位点的T/C多态性与2型糖尿病的发病具有一定程度的相关性;β3-AR基因第一外显子第190位点的T/C多态性与原发性高血压的发病无明显相关性;β3-AR基因第一外显子第190位点的T/T可能是2型糖尿病的保护性基因。 Objective To detect the relationship between genetic polymorphism of beta 3 adrenergic receptor gent and type 2 Diabetic and Essential Hypertension. Methods Polymorphism of the 190^th site T/C of β3- ARgene was detee ted by PCR-restretion fragment length polymorphism in patients with Type 2 Diabetic (n = 74) , patients with Essential Hypertension(n=82) and normal controls (n=108).Results The 190th site of β3-ARgene the frequencies of TT,TC and CC genotype was 68. 92%, 17. 57% and 13.51%in patients with Type 2 Diabetic, while 82. 93%, 13. 41 % and 3.66% in patients with Essential Hypertension and 84.26%,12.96% and 2.78% in normal controls. There is a signifi cant difference between groups of patients with Type 2 Diabetic and patients with Essential Hypertension, patients with Type 2 Diabetic and normal controls . No significant was seen in patients with Essential Hypertension and normal con trois. Conclusion The distribution frequency of β3-ARgene has something to do with morbility of Type 2 Diabetic bul has nothing to do with morbility of Essential Hypertension. Genotypes T/T inβ3-ARgene may have a protective effect against Type 2 Diabetic.
