摘要
蔗糖磷酸合成酶是植物蔗糖代谢中关键限速酶之一,为了更具体分析光响应元件ATCT-motif与分生组织特异性元件CAT-box的功能,将含有ATCT-motif和CAT-box的SPSⅢ5'侧翼序列(-1 320~-1 210),顺式作用元件SPSⅢ-AB,构建应用酵母单杂交体系的诱饵报告载体pAbAi-SPSⅢ-AB,并转化进酵母,为进一步探讨SPSⅢ基因的调控序列奠定基础.
Sucrose phosphate synthase is the key enzyme for sucrose accumulation in plant, which contains two specific domain in its promoter region, light-response element ATCT-motif and meristem-specificity element CAT-box. To concretely analyze the function of ATCT-motif and CAT- box from sucrose phosphate synthase Ⅲ in sugarcane, the fragment containing these two elements from promoter region ( - 1 320- - 1 210) was cloned and inserted into yeast one-hybrid bait vector to construct pAbAi-SPS Ⅲ-AB vector which then was transformed to yeast to screen the binding proteins. It provided a framework for further investigation on the regulation mechanism of SPS gene in sugarcane.
