黄芪药材高效液相色谱-二级管阵列检测法指纹图谱的色谱柱评价研究

摘要 目的开展黄芪药材HPLC-DAD色谱指纹图谱多色谱柱评价研究。方法色谱柱:依利特HypersilODS 2柱(A);汉邦Lichrospher C18柱(B);迪马DiamonsilTMC18柱(C);流动相:A相为乙腈-甲醇(体积比9∶1),B相为水,梯度洗脱;流速1.0 ml/min;柱温25℃;检测波长260 nm;进样量20μl。进行指纹图谱方法学考察,并进行相似度计算和统计学分析。结果黄芪药材HPLC-DAD色谱指纹图谱的方法学考察结果良好,可以标定11个共有峰。A柱和B柱色谱间相似度差异无统计学意义(P>0.05)。而C柱与A柱和B柱间的相似度差异有统计学意义(P<0.05)。从平均值可以看出,A柱和B柱之间的相似度比较一致,且均高于C柱。结论相似度计算和统计学分析可以有效评价不同色谱柱在黄芪药材HPLC-DAD指纹图谱的适用性,有利于选择合适的色谱柱,建立完善的指纹图谱评价体系。 Objective To complete column evaluation of HPLC-DAD chromatographic fingerprints of Radix as- tragali. Methods The chromatography conditions were with Hypersil ODS 2 column (A); Lichrospher C18 column (B); DiamonsilTM C18 column (C); mobile phase A: ACN-MeOH (9: 1 v/v), B: H2O, gradient elution, flow speed was 1.0 ml/min, temperature of column was 25℃ , detection wavelength 260 nm, inject volume 20 μl. The validation of the method was confirmed, and applied similarity calculation and statistics analysis. Results HPLC-DAD chromato- graphic fingerprints of Radix astragali was with better stability, precision and reproducibility, and 11 common peaks were demarcated. The results of similarity calculation showed no difference between column A and column B ( P 〉 0. 05 ) , the statistics analysis showed colunm A and B were better than C ( P 〈 0.05). Conclusion The similarity calculation and statistics analysis could effectively distinguish different colunm if HPLC-DAD chromatographic fingerprints of Radix astra- gall, which suite for the selection of column and establishment of consummate fingerprints evaluation system.
