人参皂苷Rg1抑制TGF-β1诱导肾小管上皮细胞转分化实验研究 被引量:1

摘要 目的 :通过观察人参皂苷Rg1对转化生长因子-β1(TGF-β1)诱导的大鼠肾小管上皮细胞(NRK52E)细胞转分化的影响,探讨人参皂苷Rg1在肾小管纤维化形成方面的作用。方法 :将NRK52E细胞用含10%胎牛血清的DMEM培养基培养;实验分为空白对照组、TGF-β1诱导组(5 ng/mL TGF-β1),人参皂苷Rg1与TGF-β1共同干预组(5 ng/mL TGF-β1的基础上分别加入10、20、40μg/mL的人参皂苷Rg1),培养72 h。在倒置显微镜下观察各组细胞形态的改变,并通过CCK8法检测人参皂苷Rg1对TGF-β1诱导的肾小管上皮细胞转分化的影响。结果 :TGF-β1能显著诱导大鼠肾小管上皮细胞纤维样改变,与对照组相比有明显差异(P<0.05),人参皂苷Rg1能抑制TGF-β1的作用,且其作用随浓度增加而增加。结论 :人参皂苷Rg1能抑制TGF-β1诱导的大鼠肾小管上皮细胞间充质转分化的作用,可以延缓或抑制肾间质纤维的过程。 Objective: To observe the effect of Ginsenoside Rgl on rat renal tubular epithelial- mesenchyma] transition induced by TGF-β1 , an(t to discuss the effect of Ginsenoside Rgl on tubulointerstitial fibrosis. Methods: NRK52E cells were maintained in DMEM medium supplemented with 10% fetal calf serum, and divided into control group, 5 ng/mL TGF- β1-induced group, and NRK52E co-treated with C, insenoside Rgl ( 10,20,40 μ g/mL ) and TGF-β1 ( 5 ng/mL ) . NRK52E cells were incubated for 72 hours, then light microscope was performed to observe the morphology of the cells ofeach group, and CCK8 measurement was used to detect the effect of Ginsenoside Rgl on the tubular epithelial-myofihroblast transdifferentiation induced by TGF- β 1. Hesttlt : In 5:ng/mL TGF- β 1-induced group, NRK52E cells showed fihroblast-like in morphology under light microsope, and cell proliferation was inhibited ( P〈0.05 ), when comparing with the positive control. The Ginsenoside Rgl could inhibi! the change in morphology by TGF- β 1 in a dose-dependent manner. Conclttsions : Ginsenoside Rgl could prevent TGF- β 1 inducing EMT of cuhured NRK52E cells, and retard or inhibit the process of lulmlointerstitial fibrosis.
