摘要
目的建立测定骨髓炎载药人工骨植入术后患者引流液中万古霉素含量的HPLC方法。方法以去甲万古霉素为内标,采用CAPCELL PAK C_(18)(250 mm×4.6 mm,5μm)色谱柱,流动相:0.05 moL·L^(-1)磷酸二氢钾溶液(调节pH至3.2)-乙腈(91.5:8.5),DAD检测器,检测波长280 nm,流速1 mL·min^(-1),柱温30℃,引流液经高氯酸沉淀蛋白后进样测定。结果万古霉素含量测定方法的线性范围为23.17~695.10 mg·L^(-1),r=0.9999(n=10),平均回收率102.53%,RSD=2.49%,术后第3天引流液中万古霉素浓度达峰值,累积释放量为45%。结论该方法快速简单,重复性好,适用于引流液中万古霉素浓度的检测。
AIM To establish a HPLC method for the detemaination of vancomycin in drainage tissue fluid of os- teomyelitis patients operated with microsurgical flap implants containing of vancomycin. METHODS The sample was treated by adding internal standard. A CAPCELL PAK C18(250 mm × 4.6 mm, 5 tan) column was adopted with the mo- bile phase: 0.05 moL· L^-1 potassium dihydrogen phosphate(pH 3.2)-acetonitrile( V: V = 91.5:8.5) at the flow rate of 1 mL·min^- 1 and UV detector at 280 nm. The column temperature was 30℃. Drainage tissue fluid samples were prepared by adding pewhloric acid to deposit the protain prior to analysis. RESULTS The linear range was 23.17 - 695.10 mg· L^- 1, r = 0.999 9 ( n = 10). The average recovery rate was 102.53 %, RSD = 2.49 %. The concentration of vancomycin in the drainage tissue fluid at the third day after operation got the peak value, and the cumulative release rate was 45 %. CONCLUSION The optimized method is simple, accurate and specific for the determination of vancomycin in drainage tissue fluid of osteomyelitis patients.
