DDPH对心肌肥厚大鼠PCNA,TGFβ_1,Collagen Ⅲ蛋白表达水平的影响 被引量:3

摘要 目的 研究PCNA、TGFβ1、CollagenⅢ型在心肌组织的表达及DDPH的作用。方法 用部分狭窄腹主动脉方法 ,从术后第 4周开始 ,ig不同剂量的DDPH (2 5、5 0mg·kg-1·d-1)持续 8wk。用免疫组织化学方法检测PCNA、TGFβ1、CollagenⅢ蛋白表达的变化。 结果 术后 12wk ,发现肥厚组心肌组织PCNA、TGFβ1、CollagenⅢ表达水平分别是对照组的 1 5、1 4和 2 4倍。而DDPH两个给药组蛋白表达水平明显低于肥厚组 ,但仍高于对照组。结论 DDPH对大鼠腹主动脉部分狭窄所致心肌肥厚PCNA、TGFβ1、Col lagenⅢ表达水平的增加有一定逆转作用。 AIM To investigate changes in cardiac transforming growth factor β 1 (TGFβ 1)、PCNA and Collagen type Ⅲ protein levels in cardiac hypertrophy caused by partly narrowing abdominal aorta. METHODS Rats of cardiac hypertrophy were divided into four groups randomly: control;Model;DDPH(25 mg·kg -1 ·d -1 );DDPH(50 mg·kg -1 ·d -1 ) 4 wk after operation, the rats were given drugs for 8 wk;heart weight was measured and the protein levels of PCNA,TGFβ 1,CollagenⅢ were determined by immunohistochemistry. RESULTS Protein expression levels of PCNA, TGFβ 1,CollagenⅢ in model group were more than those in control group. Protein expressions in two DDPH groups were significantly lower than in model group. CONCLUSION DDPH can reduce the protein expression of PCNA、TGFβ 1、Collagen Ⅲ in rats with cardiac hypertrophy.
