摘要
目的肝细胞肝癌(hepatocellular carcinoma,HCC)是我国常见恶性肿瘤。作为其主要标志分子的甲胎蛋白(α-fetoprotein,AFP)表达具有一定局限性,临床需要联合其他标志分子以提高诊断准确性。文中旨在探讨若干候选生物标志物在中国人源HCC细胞和正常肝细胞中的表达和定位。方法采用直接或间接免疫荧光染色方法,探讨角蛋白(cytokeratin,CK)、去唾液酸糖蛋白受体1(asialoglycoprotein receptor1,ASGR1)和ASGR2、细胞表面分子(GPC3、CD90、CD133和EpCAM)等7种主要HCC表面标志分子在Bel-7402、Bel-7404和SMMC-7721等人HCC及正常肝细胞HL-7702的表达与定位情况。结果 CK、ASGR1和ASGR2在所有的肝来源细胞均表达;GPC3仅在HCC细胞表达。CD90、CD133和EpCAM仅在少量肝肿瘤干细胞(liver cancer stem cells,LCSCs)表达。结论 CK、ASGR1和ASGR2为肝细胞标志,GPC3是HCC相关标志;CD90、CD133和EpCAM仅在极少数肝肿瘤干细胞表达。
Objective Hepatocellnlar carcinoma (HCC) is one of the most common malignant tumors with a high incidence rate in China. Alpha-fetaprotein (AFP) has some limitations in the diagnosis of HCC, so its combination with other hiomarkers could improve the diagnostic accuracy. The aim of this study is to explore the expression and location of some candidate biomarkers in Chi- nese HCC cell lines and normal hepatocytes. Methods Using direct or indirect immunofluorescent staining, we studied the expressions and locations of the candidate biomarkers CK, ASGRt, ASGR2, GPC3, CD90, CD133 and EpCAM in Bel-7402, Bel-7404, SMMC-7721 in human HCC and HL-7702 in normal hepatocytes. Results CK, ASGR1 and ASGR2 were detected in all the cell lines, while GPC3 merely in HCC cells. CD90, CD133 and EpCAM were expressed in only a few of liver cancer stem cells. Conclusion CK, ASGR1 and ASGR2 are common biomarkers of hepatocytes, while GPC3 is an HCC-related biomarker. CD90, CD133 and EpCAM are expressed in a small fraction of liver cancer stem cells.
