摘要
目的 建立自体静脉移植模型 ,并观察其平滑肌细胞 ( SMC)增殖和内膜增厚的时空变化 .方法 用“Cuff”技术将兔自体颈外静脉移植入同侧颈动脉系统 ,观察 PCNA和 α-actin表达 ,并对自体移植静脉管腔、内膜、中膜面积定量分析 .结果 自体静脉移植后扩张 ,2 wk内最明显 ,同时 SMC增殖和迁移 ,1~ 2 wk达高峰 ,PCNA和 α- actin表达也明显增高 .结论 该模型设计科学 ,成功率高 .SMC增殖从术后 3 d即已开始 ,1~ 2 wk达高峰 ,是抑制细胞增殖的最佳时期 ,增生内膜主要是由中膜
AIM To establish an animal model of autologous vein graft and to observe the proliferation of SMC and the hyperplasia of neointimal of the grafts. METHODS With the “Cuff” technique, the external jugular veins of New Zealand rabbits were implanted into the external carotid of the same side to observe the expression of PCNA and αactin in SMC and to assay the area of vessel lumen, intima and media quantitatively. RESULTS The distension of the AVG occurred after implantation, especially apparently in two weeks. Meanwhile the SMC proliferated and migrated and the peak proliferation occurred in 1~2 weeks. The expression level of PCNA and αactin increased. CONCLUSION The model was planned scientifically with a high successful rate. Proliferation began on the 3rd day after vein grafts implantation, and the peak proliferation occurred in 1~2 weeks, which was the most favourable phase to inhibit the cell proliferation. The intimal hyperplasia might be induced mainly by the proliferation and migration of the media SMC.
