摘要
目的 探讨体外循环 ( CPB)过程中肿瘤坏死因子 - α( TNF-α)在家兔胃粘膜中的表达及细胞定位 .方法 采用家兔 CPB模型 ,运用反转录 PCR及 EL ISA的方法 ,结合血浆中TNF- α、内毒素浓度的变化 ,探讨了 TNF- α m RNA的表达及细胞定位的规律 .结果 1CPB过程中 TNF-α m RNA在胃肠道的相对表达量较对照组明显增高 . 2 CPB过程中胃肠道组织中的 TNF-α呈不同程度的增高 ,而且胃肠道组织中TNF- α浓度高于血浆中的浓度 . 3CPB过程中血浆内毒素浓度较对照组明显增高 ( P<0 .0 1) ,其增高程度与胃肠道组织中TNF-α的释放及基因表达相一致 .结论 CPB中 ,组织及血浆中内毒素和 TNF- α明显增高 ,胃窦部粘膜 TNF- α m RNA的相对表达量增高可能是
AIM To elucidate the kinetics of expression and release of the tumor necrosis factorα in the gastric mucosal, mechanism of the production, and the role in complications of the open heart operations. METHODS Using CPB in rabbit, together with RTPCR and ELISA etc, the expression of TNFα mRNA, and the release of TNFα in the gastrointestine, the relationships with organ damage and with exdotoxin translocation in tissues were investigated systematically. RESULTS ①The expression of TNFα mRNA in the gastrointestinal trunk could be successively increased during CPB. ② TNFα level in gastrointestine was elevated but in different degrees during CPB. Levels and duration of TNFα in tissues were greater than those in plasma. ③Endotoxin levels in plasma were markedly increased during CPB, which was parallel to the gene expression and release of TNFα in the gastrointestinal trunk. CONCLUSION The release of TNFα in tissue and plasma is related to endotoxin translocation in tissue during CPB. It might play an important role in complications of the open heart operations.
