摘要
为提高目前高致病性蓝耳病(HR-PRRS)灭活疫苗的免疫效果,将经Marc-145细胞培养收获的HP-PRRS病毒原液超滤浓缩50倍,再经灭活后用Sepharose 4 Fast Flow分子筛柱层析,收集完整的病毒粒子,加油佐剂制成纯化HP-PRRS灭活疫苗。纯化后疫苗的攻毒保护率达到4/5以上,血清抗体检测阳转率达86%以上,均高于同批病毒液制成的未纯化灭活疫苗,且差异显著。试验证明,HR-PRRS病毒液经浓缩纯化制成疫苗后,增强了免疫效果。
To improve purification and protective potency of HP- PRRS inactivated vaccine, HP- PRRS vires multiplying and collected from Marc - 145 cells was concentrated 50 times and then inactivated. Complete virion is separated and collected by chromatography with Sepharose 4 Fast Flow. Oil adjuvant was added to prepare purified in- activated vaccine. Protective potency of purified vaccine is above 4/5 and positive conversion rate of antibody is over 86 % that all higher than that of unpurified vaccine. The differences are significant. The experiment illustrated that vaccines through concentrating and purifying can enhanced immune efficacy and non- viral protein can be removed.
