Let-7对施万细胞凋亡的调节作用

摘要 目的:探讨Let-7对施万细胞凋亡的调控作用及其发挥作用的靶基因。方法:采用Annexin V/PI双染色法检测Let-7对H2O2刺激的原代培养的施万细胞凋亡的调控作用,双荧光素酶报告基因系统检测Let-7对白介素10的3’UTR的直接靶向作用。结果:Let-7能够抑制H2O2刺激的原代培养的施万细胞的凋亡,并可直接作用于白介素10基因的3’UTR。结论:Let-7可以直接靶向IL10的3’UTR,从而通过下调靶基因IL10的表达调控施万细胞的凋亡。 Objective: To investigate the regulatory role of Let-7 in Schwann cell apoptosis and the target gene of Let-7. Methods: Annexin V/PI double-staining assays was used to identify the regulatory role of the Let-7 in the apoptosis of H2O2-stimulated primary Schwann cells and dual luciferase reportor system was used to determine the target role of Let-7 on IL10 3'UTR. Results: Let-7 inhibits the apoptosis of H202-stimulated primary cultured Schwann cells and targets the 3 'UTR of IL10 gene directly. Conclusion:This study suggests that Let-7 inhibits the Schwann cell apoptosis by regulating the expression of its target gene IL10.
