摘要
以壳聚糖和氯乙酸为原料,自制水溶性的羧甲基壳聚糖(CMCTS)。采用纸色谱和红外光谱对其进行表征并测定其水溶性和取代度。在pH=11.0的NaHCO_3-NaOH缓冲溶液中,羧甲基壳聚糖对荧光素的荧光强度具有明显的猝灭作用,且在一定浓度范围内,其猝灭程度与加入的壳聚糖浓度成线性关系,据此建立了一种新的测定壳聚糖含量的荧光猝灭法。考察了pH值、缓冲溶液类型及其用量和反应时间对荧光强度的影响。结果表明:该法的回归方程为△F=-0.097 04+0.714 68 p,r=0.999 12。检出限为2.49 mg/L。
The water-miscible Carboxymethyl chitosan(CMCTS) was synthesized with chitosan and chloroethanoic acid as trum, and, its water tensity of fluorescein raw materials. It was characterized by paper chromatography and infrared spec solubility and substituting degree were discussed as well. The fluorescence in could be quenched obviously by CMCTS in NaHCOa-NaOH buffer solution (pH=11. 0), while, at a certain concentra for the determination the quenching degree was in linear relationship with the concentration of chitosan tion range. A sensitive fluorescence quenching method was therefore developed of chitosan. The influences to the fluorescence intensity were discussed by chec-king pH value, the type and amount of buffer solution and reaction time. The results showed that the linear equation of the method was △IF =-0. 097 04+0. 714 7ρ,r=0. 999 12 and the detection limit was 2.49 mg/L.
