幼龄SD大鼠肺炎克雷伯杆菌肺炎模型的建立及炎症改变的研究 被引量:3

摘要 目的:探索建立幼年大鼠肺炎克雷伯杆菌肺炎模型的方法。观察模型的临床表现和C反应蛋白(CRP)、皮质醇、白介素-2(IL-2)、白介素-12(IL-12)、血清单核细胞趋化蛋白-1(MCP-1)水平等。方法:48只4周龄SD大鼠随机分为对照组、模型组,以上各组按观察日龄再分成3小组。每小组8只,一定浓度的细菌菌液,对照组接种生理盐水。各小组于第3、5、7 d取血清测CRP、皮质醇、IL-2、IL-12、MCP-1等。结果:建立成功的模型组IL-2、IL-12、CRP、MCP-1、皮质醇升高;模型组IL-2、IL-12、CRP、MCP-1水平随着时间延长有升高趋势,皮质醇则下降,对照各小组间无差异。结论:本实验建立的肺炎模型更简便实用;肺炎模型CRP、皮质醇、IL-2、IL-12、MCP-1改变,能够作为判断大鼠肺炎病情改变的指标。 Objective :To find out an approach to establishing the animal models in immature Sprague-Dawley (SD) rats with pneumonia induced by the Klebsiella pneumoniae bacterium for observing the clinical pictures and the level changes of C-reactive protein (CRP) , cortisol, intedeukin-2 ( IL-2 ) , interleukin-12 ( IL-12 ) and monoeyte chemotaetic protein-1 ( MCP-1 ) in the models. Methods : Forty-eight SD rats aged 4 weeks were evenly randomized into groups of controls and models, and the two groups were then subgrouped into three(n = 8 for each) , respectively, in com- pliance with days to observe. Certain dosage of Friedlander's bacillus was administered in the murine models via tracheal intubation and the controls were managed with normal saline. The blood sera were obtained in each group by the 3rd, 5th and 7th day to detect the levels of CRP, cortisol, IL-2, IL-12 and MCP-1. Results: The murine models were successfully established. The levels of CRP, cortisol, IL-2, IL-12 and MCP-I were found elevated and increased over time, whereas cortisol level was de- creased with time extension. No difference was seen in the control groups. Conclusion:The approach to establishment of an animal model seems simple and practical for experimental purpose. The level changes in CRP, cortisol, IL-2, IL-12 and MCP-1 may serve as indicators to assay the pro- gressive conditions of Klebsiella pneumonia in clinic.
