正交设计优化以沼液为基质的枯草芽孢杆菌S37最优发酵培养基 被引量:2

Optimization of Fermentation Medium of Bacillus subtilisS37 Based on Biogas Liquid by the Orthogonal Design Method

摘要 为了获得以沼液为基质的枯草芽孢杆菌S37的最优发酵培养基,以S37生物量含量为评价指标,采用单因子试验、正交试验对枯草芽孢杆菌S37的发酵培养基进行单因素多水平和多因素多水平考察。结果表明:最优发酵培养基为MgSO4·7H2O 1.6g/L、KNO30.6g/L、CaCO32.3g/L、玉米粉2g/L、KH2PO42.6g/L、豆粕3.0g/L。最佳培养基条件下,枯草芽孢杆菌生物量吸光值达到1.005,其OD600值比优化前的0.275提高了2.65倍。筛选出了发酵枯草芽孢杆菌的最佳培养基配方,为以沼液为基质发酵枯草芽孢杆菌S37工业化生产提供了技术依据。 To get the best fermentation medium of Bacillus subtilis S37 with biogas liquid for matrix fermentation,the components of biogas liquid medium for growth of B.subtilis S37 were investigated from single-factors and poly-levels and poly-factors and poly-levels by single factor experiment and orthogonal design experiment,and the bacteria biomass content of S37 was chosen as the experiment index.The results showed that the optimized conditions were:MgSO4·7H2O 1.6 g/L,KNO3 0.6 g/L,CaCO3 2.3 g/L,corn flour 2 g/L,KH2PO4 2.6 g/L,soybean meal 3.0 g/L.As a result,the fermented liquid absorbency effect could reach 1.005,2.65 times higher than that of initial medium before optimization.The research selects optimum culture medium of B.subtilis S37 with biogas liquid for matrix fermentation,which provides technical guidance for industrialization of its production.
