当归补血汤对创伤小鼠血β-内啡肽和皮质醇的影响与免疫保护作用

The influence of Dangguibuxue decoction(当归补血汤) on immunity and the production of β-endorphin and cortisol in traumatized mice

目的 :研究当归补血汤对创伤应激后小鼠的免疫调节作用 ,探讨其对应激激素释放的影响。方法 :昆明种小鼠随机分为中药治疗组、对照组、正常组。中药治疗组及对照组用落体撞击法造成小鼠双下肢股骨一次性闭合性骨折 ,对照组中取 7只于伤后 1小时放血处死后取脾行免疫功能检测。中药治疗组及对照组每日 2次灌胃注入中药或生理盐水 ,并分别于伤后第 1、2、5日早 8:30取血 ,用放射免疫法测定血β内啡肽 (β ET)、皮质醇 (Cor)含量 ,3组均于第 5日取血后取脾测定淋巴细胞对植物血凝素 (PHA)的增殖反应。结果 :中药治疗组脾淋巴细胞对 PHA的增殖反应性 (A值 )与对照组比较明显提高 ,β ET、Cor明显降低 (P<0 .0 5或 P<0 .0 1) ,且治疗组伤肢无坏死发生。结论 :创伤应激后 ,当归补血汤治疗可提高脾淋巴细胞增殖反应性 ,其作用可能与抑制应激后血中 β ET、Cor水平上升有关。

Objective:To elucidate the modulative activitis of Dangguibuxue decoction (DGBX,当归补血汤) on immunity and the production of βendorphin (βET) and cortisol (Cor) in traumatized mice.Methods:Kunming mice were divided randomly into DGBX treated(Ⅰ),traumatized control(Ⅱ) and normal(Ⅲ) groups.The mice of Ⅰ and Ⅱ groups were made closed fracture on two posterior legs by a method of dropping substance strike.After one hour,seven mice of Ⅱ group were killed by hemospasia and the proliferative activity of spleen T cells was measured by MTT assay.The mice of Ⅰ and Ⅱ groups were treated with DGBX or normal saline respectively.Seven mice of each group were sampled at 8:30 of the first,second and fifth day after traumatized,and the βET and Cor concentrations of plasma were measured by radioimmunoassay (RIA).The proliferative activity of spleen T cells was measured at the fifth day.Results:The proliferative activity of spleen T cells was significantly higher than that of traumatized control group.The concentration of βET and Cor were significantly lower than those of traumatized control.There was no necrosis at the traumatized legs of DGBX treated mice.Conclusions:DGBX has the ability to improve the proliferative activity of spleen T cells after traumatized and the immunomodulative activity of DGBX may be correlated with the suppression of βET and Cor production.
