摘要
选用不同外植体对邓恩桉组培快繁的初代培养、继代培养、生根培养等各阶段条件进行研究。结果为:以截杆萌发的邓恩桉幼嫩芽条为外植体,采用F+0.5ug·L~6-1 BA+0.3mg·L-1 IBA培养基,有利于外植体的不定芽诱导以及初期的继代扩大培养,增殖系数达3.68;以改良F+0.25mg·L~6-BA+0.25—0.5mg·L-1 IBA+20mg·L-1、rC+10mg·L。VB2为培养基,培养初期进行黑暗处理10d,增殖系数达3.26,小苗高度达1~2.5cm,适宜用于进一步的生根培养;以DR为邓恩桉生根基本培养基并添加蔗糖40g·L-1+1.0mg·L-1AST1号生根粉,生根诱导率为84.67%。
Different explants from Eucalyptus dunnii were selected to study the optimal conditions for growth at the stages of initial culture, subculture and rooting. The results indicated that adventitious buds from shoot segments of Euca/yptus durm/i could be induced, as well as propagated at the initial subculture stage on a medi- um of F + 0.5 nag·L -1 6 - BA + 0.3 mg·L -11BA, with the multiplication coefficient being 3.68. Im- proved medium ofF +0.25 mg·L--6-BA +0.25-0.5 mg· L-1 IBA+ 20mg·L-VC + 10mg· L-1 VB2 along with dark treatment for 10 days proved appropriate for sprout explants, with the multiplication coeffi- cient and height of seedling being 3.26 and 1 ~ 2.5 cm, respectively. Medium of DR supplemented with 40 g·L-1 sugar and 1.0 mg· L -1 1 # ABT rooting powder was used for Eucalyptus dunnii rooting culture, with rooting rate of up to 84.7%.
