大鼠创伤性脑损伤后高压氧治疗的时间窗研究 被引量:1

摘要 目的观察不同开始时间和不同次数高压氧(HBO)治疗对创伤性脑损伤(TBI)大鼠神经损伤的疗效,明确HBO治疗的时间窗和有效的治疗次数。方法采用Feeney法制备TBI模型,分别在致伤3、6、12、24、48、72 h后开始HBO治疗,观察治疗后大鼠神经行为学、海马结构形态学的变化,确定HBO治疗的有效时间窗和治疗次数。结果治疗组伤后3、6、12 h进行1次HBO治疗,大鼠海马CA2、CA3区锥体细胞形态及神经行为学评分显著改善,细胞丢失显著减少,其中3、6 h的作用最佳(均P<0.01)。而伤后24、48、72 h进行1次HBO治疗,与对照组比较,差异均无统计学意义(均P>0.05)。伤后6 h,进行1、3、5次HBO治疗,各次治疗后在神经行为学、海马结构形态学方面的差异均无统计学意义(均P>0.05)。但伤后24 h,进行1、3、5次HBO治疗后,3、5次治疗效果明显好于1次治疗(均P<0.05)。结论大鼠TBI后6 h内起行HBO治疗效果显著,增加治疗次数有助于增加疗效。 Objective Observe effects of hyperbaric oxygen (HBO) therapies implemented at diffe- rent time points and by different times after rat traumatic brain injury( TBI), to determine effective time window and effective therapy time of HBO therapy. Methods Feeney method was adopt to prepare TBI model, HBO therapy were implement respectively after 3, 6, 12, 24, 48 and 72 h of injury, change of rat neurological behavior and hippocampus structural morphology were observe after therapies implemented at different time points and by different times to determine effective time window and effective therapy time of HBO therapy. Results Implement ation of HBO therapy once within 3, 6 and 12 h after therapy group's injury could remarkably improve rat neurological behavior score and pyramidal cell morphology of hippocampus CA2 and CA3 areas and could reduce cell loss, especially within 3 and 6 h ( P 〈 0.01 ). HBO therapy implemented within 24, 48 and 72 h after injury has no statistical difference with control group ( all P 〉 O. 05 ). HBO therapies implemented for 1,3 and 5 times within 6 h of injury have no statis- tical difference on neurological behavior and hippocampus Structural morphology ( all P 〉 0.05 ). Within 24 h of injury, therapeutic effects of therapies implemented for 3 and 5 times are more excellent than that implemented once ( all P 〈 0.05). Conclusion HBO therapy implemented within 6 h after rat TBI will take remarkable effect, and increment of therapy time is beneficial for increasing the therapeutic effect.
