摘要
目的探讨胃癌组织中叉头蛋白3(Foxp3)蛋白表达与胃癌腹膜转移的关系。方法应用鼠抗人Foxp3单克隆抗体和免疫组织化学SP染色法检测65例同期伴腹膜转移的胃癌患者的癌旁正常胃黏膜、胃癌原发灶、胃癌腹膜转移灶及淋巴结转移灶中Foxp3蛋白的表达水平。结果胃癌原发灶、胃癌腹膜转移灶及淋巴结转移灶组织中Foxp3蛋白呈强表达,阳性表达率分别为78.5%(51/65)、80.0%(52/65)和88.6%(39/44);胃癌旁正常胃黏膜组织中Foxp3蛋白表达很弱,阳性表达率为20.0%(13/65)。正常胃黏膜组织中Foxp3蛋白阳性表达率显著低于胃癌原发灶、腹膜转移灶及淋巴结转移灶组织(P值均<0.05),后3者间的差异无统计学意义(P值均>0.05)。Foxp3蛋白的阳性表达率与年龄、性别不相关(P值均>0.05),而与组织分化程度呈负相关(r=-0.301,P=0.023),与有无淋巴结转移呈正相关(r=0.358,P=0.004)。结论胃癌病灶中的Foxp3蛋白可能成为术前判断胃癌是否发生腹膜转移及判断预后的生物学指标。
Objective To investigate the relationship between the expression of Foxp3 protein and peritoneal metastasis in gastric cancer. Methods The expressions of Foxp3 was detected in matching samples including primary gastric cancer lesions, lymph node metastases, peritoneal metastases and normal tissues by immunohistochemistry. The specimens were obtained from 65 patients who had synchronous peritoneal metastasis from gastric cancer. Results The expression of Foxp3 was 78.5% (51/65), 80.0% (52/65), 88.6% (39/44) and 20.0% (13/65) in the gastric cancer lesions, peritoneal metastases, lymph node metastases and normal tissures, respectively. As compared with normal tissues, the expression of Foxp3 in the gastric carcinoma lesions, peritoneal metastases and lymph node metastases was significantly increased (all P〈0.05) but there were no significant differences among the gastric cancer lesions, peritoneal metastases and lymph node metastases (all P,〉0.05). The expression of Foxp3 was not correlated with age and sex of the patients (both P〉0.05), whereas it was negatively correlated with histological differentiation ( r = - 0. 301, P = 0. 023) and positively correlated with lymph node metastases (r = 0. 358, P = 0. 004). Conclusion The expression of Foxp3 in gastric cancer lesions can be used as a biological marker of peritoneal metastasis from gastric cancer before operation and patient prognosis.
