尿中同型胱氨酸纸层析法鉴定及其半定量

IDENTIFICATION AND SEMIQUANTITATION ANALYSIS OF URINARY HOMOCYSTINE WITH PAPER CHROMATOGRAPHY

本文以Whatman 3MM滤纸作小纸快速单向层析,经硝普银显色。根据Rf值和显色的差异性鉴定同型胱氨酸。平行层析系列标准液,比较色泽深浅推断24h尿液内的同型胱氨酸含量。方法简单易行,是鉴定和半定量同型胱氨酸的理想方法之一。

Homocystinuria is a biochemical disorder caused by deficiency of cystathionine β-synthase. A rapid one-dimensio nal paper chroma-tography was carried out with Whatman 3 MM and then stained by silver nitroprusside. The homocystine was identificated according to characteristic colors. At the same time a series of standard solutions were made simultaneously on the same paper. The characteristic color intensity of patient urinary chromatogram pattern was compared with that of standards. Homocystine amounts in 24-hour urine were calculated. The method proved to be simple and easy, in the identif icat ing and semiquantitating of homocystine.
