摘要
目的建立超高效液相色谱(ultra performance liquid chromatography,UPLC)快速测定旋覆花属植物中2种倍半萜内酯的含量.方法仪器为Waters ACQUITY Ultra Performance LCTM超高效液相色谱仪,带有二极管阵列检测器,检测波长205 nm,色谱柱为Waters ACQUITY UPLC BEH C18(100 mm×2.1 mm,1.7μm),以乙腈-水为流动相,梯度洗脱,流速0.4 mL/min,柱温:25℃.结果 2种倍半萜内酯在7 min内完成测定.结论该方法简单、准确可靠、重复性良好,可用于旋覆花属植物中2种倍半萜内酯含量的快速测定.
Objective To determine the contents of two active sesquiterpenes in Inula plants with UPLC. Methods The equipment was Waters ACQUITY Ultra Performance LCTM UPLC. Detection wavelength was 205 nm. The column was Waters ACQUITY UPLC BEH C18 (100 mm 2.1 mm, 1.7 μm). Mobile phase: A was acetonitrile and B was water with gradient elution. Flow speed was 0.4 mL/min, and the temperature of column was 25℃. Result Two sesquiterpenes (inulicin and ivangustin) were separated at baseline within 7 min. Conclusion The method was simple and accurate with good reproducibility, can be used to determine quickly the contents of two sesquiterpenes in Inula plants.
