摘要
目的:研究复方丹参片对侧脑室注射Aβ25-35诱导所致痴呆小鼠模型的学习记忆功能障碍的改善作用及对RACK1表达的影响。方法:150只雄性昆明种小鼠经水迷宫实验筛选后,随机均分成假手术组、模型组、复方丹参片高、低剂量组和石杉碱甲组(n=10),侧脑室注射凝聚态Aβ25-35 10μl建立老年痴呆模型,Morris水迷宫测定学习记忆能力,HE染色观察海马内神经元病理改变,ELISA法检测脑组织中IL-6,TNF-α及BDNF,RACK1的含量变化。结果 :与假手术组相比,模型组小鼠水迷宫逃避潜伏期明显延长,且脑组织中IL-6,TNF-α表达显著增加,BDNF与RACK1的表达下降。而复方丹参片0.405 g/kg(低剂量)至0.810 g/kg(高剂量)均能在一定程度上缩短痴呆小鼠的逃避潜伏期,降低脑组织中炎症因子表达,提高BDNF与RACK1蛋白的含量。其中以高剂量治疗效果更为明显(P<0.05)。结论 :复方丹参片可能通过调节Aβ25-35诱导所致的老年痴呆模型小鼠脑组织中炎症因子、BDNF与RACK1的表达,从而改善痴呆小鼠的学习记忆功能障碍。
To study the effects of Compound Danshen Tables on behaviors and RACK1 in Alzheimer's disease (AD) mice induced by Aβ25-35. Methods: 150 male KM mice selected by water maze were randomly divided into sham operation group, model group, high and low dose Compound Danshen Tables group and Huperzine A group, (n = 10). AD model was induced by injecting lateral ventricle with 10 p.] A[325-35, learning and memory capability was observed with Morris water maze. hippocampal neurons injuries was observed by HE stain and expression of IL-6, TNF-a ,RACK1 and BDNF in brain tissue were tested by enzyme-linked immune sorbent assay (ELISA). Results: Com- pared to sham operation group, model group had ]onger escape latency, increased the expression of IL-6, TNF-a and decreased the expres- sion of BDNF, RACK-1 (P 〈0.01, P 〈 O. 05). Compound Danshen Tables could significantly shorten escape latency, decrease the expres- sion of IL-6, TNF-α and increase the expression of BDNF, RACK-1 of AD mice( P 〈0.05 ). Conclusions: Compound Danshen Tables can adjust the inflammatory factors, BDNF, RACK-1 expression in the AD model mice and improve the learning and memory impairment.
