碧血胶囊对大鼠急性心肌梗死模型心功能和血流动力学的影响 被引量:5

Effects of Bixue Capsule on cardiac function and hemodynamics in rat myocardial infarct model

摘要 目的:观察碧血胶囊对大鼠急性心肌梗塞(AMI)后血流动力学变化及心功能的影响,探讨其预防AMI的机制。方法:结扎大鼠冠状动脉左前降支造成AMI模型,成年雄性SD大鼠50只随机分为6组:假手术组、急性心肌梗塞模型(AMI)组、通心络胶囊组、碧血胶囊高、低剂量组、地尔硫卓组。观察药物对各组大鼠心率(HR)、左心室收缩峰压均值(LVSP)、左心室舒张压均值(LVDP)、左心室收缩压最大上升速率(+dp/dtmax)、左心室舒张压最大下降速率(-dp/dtmax)、左心室开始收缩至达到室内压最大上升速率时间(t-dp/dtmax)、心律失常、血清游离脂肪酸(FFA)和天冬氨酸氨基转移酶(AST)的影响。结果:大鼠AMI后,HR、mlVSP、±dp/dtmax明显降低,LVDP,AST明显升高(P<0.05),并且随着缺血时间的延长,各指标呈逐渐恢复的趋势。与AMI组相比,碧血胶囊高(968mg/kg,结扎后120分钟)、低剂量组(484mg/kg,结扎时)可显著提高LVSP,碧血胶囊高(结扎时及结扎后120分钟时)、低剂量组(结扎后30分钟)可显著提高+dp/dtmax,碧血胶囊高(结扎时及结扎后30、120分钟时)可显著提高-dp/dtmax(P<0.05),同时碧血胶囊高、低剂量组均能降低血清AST及心律失常的发生次数(P<0.05)。结论:碧血胶囊能显著改善左室收缩和舒张功能,减轻大鼠心肌缺血后心功能的下降和降低血清AST含量,对AMI后血流动力学改变有明显的改善作用,这可能是其保护心肌梗死的机制之一。 To investigate the protected effects of Bixue capsules on cardiac function and hemodynamies in rats acute myocardial infarc- tion (AMI) model and to discuss its mechanism. Methods: Rat AMI model was constructed by ligating left anterior descending coronary ar- tery and the rats were randomly divided into shame group, acute myocardial infarct model (AMI) group and the low and high dosages of Bix- ue capsule groups, Tongxinluo capsule control group and Dierliuzhuo control group. We observed the cardiac function index such as heart rate (HR), left ventricular systulic( L^SP), left ventricular diastdic blood pressure (LVDP), maximal rising and failing rate of ventricular pres- sure( + dp/dtmax ), the time from beginning of left ventrieular systolic to maximal rising rate of ventricular pressure (t-dp/dt), arrhythmia and serum FFA and AST. Results: After AMI, the level of HR, LVSP and + dp/dtmax were lower than those before AMI, the levels of mlVDP and AST increased significantly ( P 〈 0.05). Along with the prolongation of ischemia all indexes tended to normal level. Compared to AMI group, Bixue capsule treated groups (high doses that is 965mg/kg , at the time of the 120 minutes after ligation and low doses treated group( that is 484 mg/kg, at the time of ligation) can significantly increased LVSP, Bixue capsule high doses treated groups(at the time of ligation and the 120 minutes after ligation ) and low doses treated groups( at the 30 minutes after ligation ) can significantly increased + dp/ dt max , Bixue capsule high doses treated groups( at the time of ligation and the 30,120 minutes after ligation ) can significantly increased - dp/dt max ( P 〈 0.05 ) . at the same time Bixuc capsule treated groups can significantly decreased AST in the serum and the happening of ar- rhythmia (P 〈 0.05 ). Conclusion: By improvfia~ the compliance of ventricular wall, Bixue capsule .treated groups can mitigate the decline of cardiac function and reduce the content of AST in the serum, at the same time l it significantly improve cardiac systolic and diastolic function after AMI, which maybe is one of the mechanism of Bixue capsule treated AML
