目的:探讨玄莲消癖口服液对乳腺增生模型大鼠的影响,为其临床应用提供实验依据。方法:取90只未孕雌性大鼠分为6组:正常对照组、模型组、消核片对照组、玄莲消癖口服液高、中、低三个剂量组,每组15只动物。正常对照组隔日腋下注射生理盐水,其余各组均隔日腋下注射雌二醇,共10次,复制乳腺增生病理模型。于造模的同时给予受试药物,正常对照组给予等量生理盐水。末次给药后采血测定雌二醇(E2)和孕酮(P)含量;用打孔器取第二对乳房做病理切片,观察病理形态学变化;取第三对乳房测定其体积(容积法测量)。同时于给药前、给药后均测量所有动物胸部第一对乳房直径。结果:玄莲消癖口服液20～40g/kg可明显改善乳腺组织增生性病理形态学变化其主要表现:明显减少乳腺小叶数和腺泡数,显著减小乳腺体积、腺泡腔直径及乳房直径;同时还能显著降低雌二醇水平,明显升高孕酮水平,从而明显降低E2/P值。结论:玄莲消癖口服液对乳腺增生模型大鼠的病理形态、激素水平均有明显的调节作用。
