Distribution and significance of three subtypes of ER in breast invasive ductal cancers

摘要 目的:探讨三种雌激素受体(estrogen receptor,ER)亚型(ERα、ERβ及雌激素G蛋白偶联受体estrogen(G protein coupled)receptor,GPER)在乳腺浸润性导管癌中的分布与意义。方法:应用免疫组织化学法检测135例石蜡包埋浸润性导管癌组织中ERα、ERβ及GPER蛋白的表达,用χ2检验等方法分析其与临床病理变量之间及相互间的关联,方差分析比较不同ER表型肿瘤诺丁汉预后指数(Nottingham prognostic index,NPI)的差异。结果:135例浸润性导管癌组织中ERα、ERβ及GPER阳性的比例分别为53.3%、64.7%及74.8%,三种ER亚型的表达无显著关联。ERα的表达与孕激素受体、人表皮生长因子受体-2显著相关,ERβ、GPER的表达与肿瘤临床病理变量没有关联。ERα-ERβ-GPER-乳腺癌平均NPI较ERα+ERβ+GPER-及ERα+ERβ+GPER+肿瘤高,差异具有统计学意义(P值分别为0.046、0.022)。结论:ERα、ERβ与GPER在浸润性导管癌中均有广泛分布,可能参与介导雌激素效应。ERα与ER及GPER相互独立,ERs表型分析对认识雌激素对乳腺癌的影响具有重要意义。 Objective: To evaluate tile distribution and significanee of tile three, subIytx-s of estrogen receptor (ER) ill manmmry invasive ductal cancer. Methods: hanlunohisthemist was used to dettxt ERct, ER and estrogen ( G protein coupled) receptor (GPER) in ptuaffin - embtdtxt ti,;u eft invasive duetal cancers frown 135 patients. The expreion of ERs was correlated to clinieopathological variahles via Chi - luase and t - test. Association analysis was prefomled among Ihe three subtypes of ER as well. In addition, mean Nottinghtun prognostic index was comparext by one - way analysis of variance among subgroups of tumor with variable ER phenotype. Results: ERa,ER and GPER were detected in 53.3%, 64.7% and 74.8% of these 135 smnples reslxeetively. However, there was no significant association among them. ERa expressicnl was carrelated to progesterone receptor and hmnan epidemlal growth lactor receptor - 2 significantly. hile, no statistical as:ciation was found between ER, as well as GPER, and mlyone of clinieopathologied variables detected. Interestingly, the mean Nottinghmn prognostic index was larger in ERa- ER- GPER- tunHrs comparing In ERa + ER + GPER- (P = 0. 046) and ERct + ER + (;PER + (P = 0.022) tnles in the analysis mnong subgrottps ff tumor with different ER phenotype. Canclusions: Any of ERa, EI mad GPER was widely distributed in invasive ductal cancers. Thus, all of them could be involved in the estrogenic response ill breast cancer. Furthermore, ERa, ER and GPER are three independent subtypes of ERrand analvsisof their p.l)enotyp would he essentitrl for the study of esrogenic effects in bast cancers.
